Methylation-specific PCR analysis in Col8A1 promoter in Creole cattle carrier of rob(1;29) Electron. J. Biotechnol.
Postiglioni,Alicia; García,Cristina B; Rincón,Gonzalo; Arruga,M. Victoria.
Robertsonian translocation (rob(1;29)) is the most frequent structural chromosomal abnormality in cattle. Heterozygous carriers have a normal phenotype but show a 3-5% decrease in fertility. Chromatin decondensation was evaluated similar to the inactive X chromosome when submitted to demethylating agent. Based on this result, and the concept that imprinted genes are essential in embryonic development, we decided to query genes located on BTA1 and BTA29 that could undergo genome imprinting. The collagen typeVIII-α1 (Col8A1) acted on extracellular matrix structural proteins. DNA bisulfite conversion and sequentiation methods were used to compare its differential methylation patterns. It was performed on eight Creole cattle DNA blood samples from normal and...

Marine Polysaccharide-Collagen Coatings on Ti6Al4V Alloy Formed by Self-Assembly ArchiMer
Norris, Karl; Mishukova, Oksana; Zykwinska, Agata; Colliec-jouault, Sylvia; Sinquin, Corinne; Koptioug, Andrei; Cuenot, Stéphane; Kerns, Jemma; Surmeneva, Maria; Surmenev, Roman; Douglas, Timothy.
Polysaccharides of marine origin are gaining interest as biomaterial components. Bacteria derived from deep-sea hydrothermal vents can produce sulfated exopolysaccharides (EPS), which can influence cell behavior. The use of such polysaccharides as components of organic, collagen fibril-based coatings on biomaterial surfaces remains unexplored. In this study, collagen fibril coatings enriched with HE800 and GY785 EPS derivatives were deposited on titanium alloy (Ti6Al4V) scaffolds produced by rapid prototyping and subjected to physicochemical and cell biological characterization. Coatings were formed by a self-assembly process whereby polysaccharides were added to acidic collagen molecule solution, followed by neutralization to induced self-assembly of...

Unusual Glycosaminoglycans from a Deep Sea Hydrothermal Bacterium Improve Fibrillar Collagen Structuring and Fibroblast Activities in Engineered Connective Tissues ArchiMer
Senni, Karim; Gueniche, Farida; Changotade, Sylvie; Septier, Dominique; Sinquin, Corinne; Ratiskol, Jacqueline; Lutomski, Didier; Godeau, Gaston; Guezennec, Jean; Colliec-jouault, Sylvia.
Biopolymers produced by marine organisms can offer useful tools for regenerative medicine. Particularly, HE800 exopolysaccharide (HE800 EPS) secreted by a deep-sea hydrothermal bacterium displays an interesting glycosaminoglycan-like feature resembling hyaluronan. Previous studies demonstrated its effectiveness to enhance in vivo bone regeneration and to support osteoblastic cell metabolism in culture. Thus, in order to assess the usefulness of this high-molecular weight polymer in tissue engineering and tissue repair, in vitro reconstructed connective tissues containing HE800 EPS were performed. We showed that this polysaccharide promotes both collagen structuring and extracellular matrix settle by dermal fibroblasts. Furthermore, from the native HE800...

Contribution Towards the Anatomy of the Esophageal Hiatus and its Relationship with the Presence of Bundles of Collagen Fibers in its Margins International Journal of Morphology
Filho,José Jeová de Oliveira; Filho,Benedito Herani; Reis,Francisco Prado; Feitosa,Vera Lúcia Correa; Aragão,José Aderval.
The aim was to detect the presence of bundles of collagen fibers in the margins of the esophageal hiatus and establish if there is any relationship of these bundles with the anatomy of the hiatus. Ten adult male cadavers, with no gross anatomical alteration caused by trauma, surgery or disease, upon the esophageal hiatus were used in the study. A piece of anatomical structure comprising the diaphragm with the esophageal hiatus and adjacent tissues was removed, dissected and 10% formol embedding. With the aid of a digital caliper, measurements of the perimeter of the esophageal hiatus were done both in the abdominal and thoracic sides. For the structural study each margin was divided in six sections. Staining techniques of Masson and Picrosirius-hematoxilin...

ULTRASTRUCTURAL ANALYSIS OF PINEALECTOMY AND LACK OF LIGHT INFLUENCE OVER COLLAGEN IN THE ENDOMETRIUM OF RATS International Journal of Morphology
Pinto de Medeiros,Juliana; Wanderley Teixeira,Valéria; Aguiar Coelho Teixeira,Álvaro; Baratella Evêncio,Liriane; Evêncio Neto,Joaquim.
Some studies have shown that pinealectomized female rats present a large concentration of collagen fibers in the endometrium. The lack of light results in morphologic alterations in the uterine lamina propria and in the muscular layer, reducing thus the uterine weight. Therefore, this work aimed to analyze ultra-structurally the influence of melatonin in the remodeling of the collagen in the endometrium, in the estrus phase, in pinealectomized adult rats or submitted to lack of light. The experimental groups: I- sham-pinealectomized maintained in a clear/dark cycle 12/12 hours, during three months; II- pinealectomized rats maintained in a clear/dark cycle 12/12 hours, for three months; III- normal rats maintained in clear/dark cycle 12/12 hours for three...

Quantitative Analysis of Collagen Fibrillar Structure in Canine Uterus Exposed to Acetate of Medroxyprogesterone Using Second Harmonic Generation Microscopy International Journal of Morphology
Salinas,Paulo; Miglino,Maria Angélica; del Sol,Mariano.
Collagen is the most abundant structural protein found in organs and is responsible for providing tissues with structure and function. In order to investigate in canine uteri the potential effect of medroxyprogesterone acetate (MPA) on the changes in collagen deposition were grouped as nulliparous (n=11), multiparous (n=11) and treated with MPA (n=11; nulliparous with two treatments; 5 mg/kg; i.m.). The amount of collagen was studied in the fold and basal regions of the endometrium and myometrium using second harmonic generation with a two-photon spectral confocal microscope, quantified using ImageJ software with a color segmentation plugin, was expressed as fraction area (%) and analyzed by ANOVA (p<0,05). No differences were observed between groups in...

Abstract Background Hemolin proteins are cell adhesion molecules from lepidopterans involved in a wide range of cell interactions concerning their adhesion properties. However, hemolin’s roles in cell proliferation and wound healing are not fully elucidated. It has been recently reported that rLosac, a recombinant hemolin from the caterpillar Lonomia obliqua, presents antiapoptotic activity and is capable of improving in vitro wound healing. Therefore, this study aimed to explore rLosac’s in vivo effects using a skin wound healing model in rats. Methods Circular full-thickness wounds in the rat dorsum skin were treated either with rLosac, or with saline (control), allowing healing by keeping the wounds occluded and moist. During the wound healing, the...

Biaxial testing of canine annulus fibrosus tissue under changing salt concentrations Anais da ABC (AABC)
Huyghe,Jacques M..
The in vivo mechanics of the annulus fibrosus of the intervertebral disc is one of biaxial rather than uniaxial loading. The material properties of the annulus are intimately linked to the osmolarity in the tissue. This paper presents biaxial relaxation experiments of canine annulus fibrosus tissue under stepwise changes of external salt concentration. The force tracings show that stresses are strongly dependent on time, salt concentration and orientation. The force tracing signature of are sponse to a change instrain, is one of a jumpin stress that relaxes partly as the new strain is maintained. The force tracing signature of a stepwise change in salt concentration is a progressive monotonous change in stress towards a new equilibrium value. Although the...

The extracellular matrix of the lung and its role in edema formation Anais da ABC (AABC)
Pelosi,Paolo; Rocco,Patricia R.M.; Negrini,Daniela; Passi,Alberto.
The extracellular matrix is composed of a three-dimensional fiber mesh filled with different macromolecules such as: collagen (mainly type I and III), elastin, glycosaminoglycans, and proteoglycans. In the lung, the extracellular matrix has several functions which provide: 1) mechanical tensile and compressive strength and elasticity, 2) low mechanical tissue compliance contributing to the maintenance of normal interstitial fluid dynamics, 3) low resistive pathway for an effective gas exchange, d) control of cell behavior by the binding of growth factors, chemokines, cytokines and the interaction with cell-surface receptors, and e) tissue repair and remodeling. Fragmentation and disorganization of extracellular matrix components comprises the protective...

Placental osseous metaplasia associated with abortion in a mare: case report Arq. Bras. Med. Vet. Zootec.
Lima,P.A.; Eckstein,C.; Ecco,R.; Santos,R.L..
ABSTRACT The aim of this report is to describe a case of marked osseous metaplasia in the placenta associated with abortion in a mare. A 19-year-old Campolina mare with a history of multiple parturitions and absence of reproductive failure, aborted a fetus with approximately six months of gestation. There were no gross changes in the fetus and the placenta. Microscopically, there was multifocal replacement of fetal connective mesenchyme of the chorionic villi with well differentiated and partially mineralized osseous tissue (confirmed by von Kossa staining). To further characterize the collagenous component of the lesion, sections of the placenta were stained with Masson’s trichrome and Picrosirius red. Osseous tissue deposition had abundance of type I...

Dermatosparaxis in two White Doper lambs in Brazil: case report Arq. Bras. Med. Vet. Zootec.
Silva,A.P.C.; Mol,J.P.S.; Carvalho Junior,C.A.; Paixão,T.A.; Santos,R.L..
Dermatosparaxis is a genetic disease that affects collagen maturation. This disease is characterized by marked impairment of the resistance of collagen fibers that leads to skin fragility, and it may affect several species. This is the first report of dermatosparaxis in sheep in Brazil. Clinically, two White Dorper lambs had multiple skin lacerations in the neck, dorsum and limbs. Transmission microscopy demonstrated irregular collagen fibers arranged in hieroglyphic shape and scanning electron microscopy demonstrated thin collagen fibrils that were not arranged in bundles as observed in the normal dermis. These findings are consistent with the diagnosis of dermatosparaxis.
